- βAverage weight loss of ~16% of body weight compared to placebo over 12β18 months
- βWeight loss was sustained at 3.5 years of follow-up in longer-term data
- βNo significant increase in serious adverse events, heart attacks, strokes, or death rates compared to placebo
- βParticipants were 3.6 times more likely to achieve the clinically meaningful 5% weight loss threshold

What Was This Study About?
Researchers reviewed 9 clinical trials involving 7,111 adults living with obesity to understand how well tirzepatide (brand name Mounjaro) works for weight loss. Tirzepatide is a weekly injection that targets two hormones β GIP and GLP-1 β to help control appetite and blood sugar.
What Did They Find?
Compared to placebo (a dummy injection), people taking tirzepatide lost an average of about 16% of their body weight over 12β18 months. For someone weighing 250 lbs, that's roughly 40 lbs of weight loss. People taking tirzepatide were also 3.6 times more likely to lose at least 5% of their body weight β a meaningful health threshold.
Importantly, this weight loss appeared to be sustained at 3.5 years in longer follow-up data, with participants maintaining around 15β16% weight reduction.
What About Safety?
Tirzepatide did increase non-serious side effects (like nausea or digestive issues) compared to placebo. However, serious side effects were similar between tirzepatide and placebo groups. There was no meaningful difference in heart attack/stroke risk (MACE) or death rates between the groups.
What About Quality of Life?
Physical quality of life scores improved slightly with tirzepatide, but researchers noted the difference may not be clinically dramatic in everyday terms.
Important Caveats
All 9 studies were funded by the drug manufacturer (Eli Lilly), which raises concerns about potential bias. Most trials focused on middle- and high-income countries, so results may not apply to all populations. Independent research is still needed.
What This Means for Diabetes Patients
Several of the trials specifically included people with type 2 diabetes or prediabetes. For these patients, meaningful weight loss from tirzepatide could help improve blood sugar control, reduce cardiovascular risk, and potentially reduce medication needs β though always discuss changes with your care team.
